摘要

目的 测定人参中的果胶含量。方法 先除去人参中的可溶性糖,然后加入果胶酶将果胶分解,再采用-3,5-二硝基水杨酸比色法对果胶含量进行测定。结果 测定人参中果胶含量的最佳条件为:供试品细粉500 mg,经80%乙醇水浴提取后,过滤,残渣加入含有果胶酶1 200 U的枸橼酸-枸橼酸三钠缓冲液50.0 mL(pH 3.5),50 ℃水浴中恒温加热6 h酶解。用DNS比色法测定酶解液中的果胶含量(以半乳糖醛酸计)。结论 本法操作方便、准确,可用于人参果胶含量的检测。

Abstract

OBJECTIVE To determine the pectin content in Panax ginseng. METHODS Soluble sugar was removed firstly, then pectinase was added for enzymolysis, and 3, 5-dinitrosalicylic acid colorimetry was employed for determination of pectin. RESULTS The optimal conditions of determination were as follows500 mg of Panax ginseng powder was extracted with 80% alcohol in water bath at 80 ℃, then filtrated, and the residue was enzymolyzed with pectinase of 1 200 u at 50 ℃ for 6 h in 50 mL of 0.1 mol·L-1 citric acid-sodium citrate buffer (pH 3.5). The galacturonic acid from degraded pectin in Panax ginseng was determined with 3, 5-dinitrosalieyfic acid colorimetry. CONCLUSION This method has been proved to be convenient and accurate, which can be applied to the determination of pectin content in Panax ginseng.
